Caution Concentrations of HEPES over 40mM may be toxic to some cell types. Preparation Note Prepared from HEPES free acid and cell culture grade water. Application HEPES is an organic zwitterionic buffering agent effective in the physiological pH range of 6.8 to 8.2 (pKa 7.55). It is typically used in cell culture at concentration between 5mM to 30 mM. This product has been qualified for use in cell culture.
Application HEPES is used in many media because it has more buffering capacity in the physiologic pH range of 7.2 - 7.4 at 37 C than sodium bicarbonate. It is commonly added at concentrations between 10 mM and 25 mM. Higher levels cause cytotoxicity. Because bicarbonate is nutritionally necessary for most cells, HEPES should be added in addition to, not in place of, sodium bicarbonate.
Other Notes 3′-Terminal labeling of RNA with T4 RNA ligase1; Isolation and analysis of nuclear RNA - component of reaction buffer, prehybridization buffer, hybridization buffer2. Easily compare specifications for HEPES products with the HEPES specification table.
Other Notes One of the best general purpose buffers available for biological research. Has been used to advantage in tissue culture, oxidative phosphorylation, protein synthesis with cell-free bacterial systems, photophosphorylation, CO2 fixation1,2; Suitable buffer for TEM studies (Transmission Electron Microscopy) as it does not affect metal substrates3; Recommended buffer for the glutamate binding assay, prevents binding to non-receptor materials4. Easily compare specifications for HEPES products with the HEPES specification table.
A HEPES that is ethanesulfonic acid in which one of the methyl hydrogens is replaced by a 4-(2-hydroxyethyl)piperazin-1-yl group. A Good's buffer substance, pKa = 7.55 at 20 degreeC.
